Deformability based Cell Sorting using Microfluidic Ratchets

Cell sorting using mechanical filters is fundamentally limited by clogging, which degrade selectivity and prevent extraction of the separated cells. To overcome this problem, we developed the microfluidic ratchet mechanism to selective transport cells based on deformability using oscillatory flow. This mechanism prevents clogging to preserve the selectivity of the filter and enables robust processing of high-density cell samples, such as whole blood. I will present the principles of this mechanism, related mechanisms for measuring cell deformability, and applications to circulating tumor cells and antimalarial drug discovery.
